{"id":1239,"date":"2023-11-15T17:05:43","date_gmt":"2023-11-15T17:05:43","guid":{"rendered":"https:\/\/wordpress.clarku.edu\/drewelllab\/?page_id=1239"},"modified":"2024-11-28T16:29:06","modified_gmt":"2024-11-28T16:29:06","slug":"molecular-mechanisms-of-transcriptional-regulation-in-drosophila-development","status":"publish","type":"page","link":"https:\/\/wordpress.clarku.edu\/drewelllab\/projects\/molecular-mechanisms-of-transcriptional-regulation-in-drosophila-development\/","title":{"rendered":"Molecular mechanisms of transcriptional regulation in Drosophila development"},"content":{"rendered":"<h3><strong>Overview<\/strong><\/h3>\n<p style=\"font-weight: 400\">The precise regulation of gene expression is critical to early embryonic development.&nbsp; This transcriptional regulation is tightly controlled by the binding and complex interactions between multiple protein transcription factors (TFs) and DNA sequences within <em>cis-<\/em>regulatory modules (CRMs or enhancers). &nbsp; We use an integrated combination of interdisciplinary approaches, including bioinformatic, mathematical modeling, biophysical and synthetic biology experiments, and transgenic reporter assays to decipher the regulatory code of the gene regulatory networks in the fruit fly model species, <em>Drosophila melanogaster<\/em>.<\/p>\n<h3><b><i>Twin of eyeless<\/i><\/b><\/h3>\n<p style=\"font-weight: 400\">The <em>Drosophila<\/em> PAX6 homolog <em>twin of eyeless<\/em> (<em>toy<\/em>) sits at the pinnacle of the genetic pathway controlling eye development, the retinal determination network.&nbsp; Expression of <em>toy <\/em>in the embryo is first detectable at cellular blastoderm stage 5 in an anterior dorsal band in the presumptive procephalic neuroectoderm, which gives rise to the primordia of the visual system and brain.&nbsp; Although several maternal and gap transcription factors (TFs) that generate positional information in the embryo have been implicated in controlling <em>toy<\/em>, the regulation of <em>toy<\/em> expression in the early embryo is currently not well characterized.&nbsp; In our studies, we adopt an integrated experimental approach utilizing bioinformatics, molecular genetic testing of putative enhancers in transgenic reporter gene assays and quantitative analysis of expression patterns in the early embryo. &nbsp;Recently, we identified two novel coacting (shadow) enhancers at the <em>toy<\/em> gene.&nbsp; In addition, we apply mathematical modeling to dissect the regulatory landscape for <em>toy <\/em>and are investigating the distinct molecular architecture of the two enhancers.<\/p>\n<p><a href=\"https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2.jpg\"><img loading=\"lazy\" decoding=\"async\" class=\"alignnone size-full wp-image-1256\" src=\"https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2.jpg\" alt=\"\" width=\"2147\" height=\"1686\" srcset=\"https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2.jpg 2147w, https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2-300x236.jpg 300w, https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2-1024x804.jpg 1024w, https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2-768x603.jpg 768w, https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2-1536x1206.jpg 1536w, https:\/\/wordpress.clarku.edu\/drewelllab\/wp-content\/uploads\/sites\/313\/2024\/11\/toy-summary-figure-2-2048x1608.jpg 2048w\" sizes=\"auto, (max-width: 2147px) 100vw, 2147px\" \/><\/a><\/p>\n<h3><strong><em>Hox<\/em> genes<\/strong><\/h3>\n<div>\n<p class=\"p1\">In the early <i>Drosophila<\/i> embryo, CRMs, under the control of TF concentration gradients, direct the spatio-temporal expression of a network of developmental genes, including the highly conserved homeotic genes in the bithorax complex (BX-C).
